Objectives: To investigate the effects of DOT™ PN injection on stem cell markers (CD31, CD34, CD166), fibroblast activation, and histological changes in the adipose and dermal layers of the face. This study aims to evaluate adipogenesis, collagen synthesis, neovascularization, and tissue regeneration to determine the therapeutic potential of DOT™ PN in aesthetic and regenerative medicine.
Introduction: Aging leads to reduced adipose tissue volume, collagen degradation, and compromised skin elasticity. Stem cell markers such as CD31, CD34, and CD166 play pivotal roles in tissue regeneration, vascularization, and adipocyte differentiation. This study explores how DOT™ PN injections influence these markers and promote fibroblast activity, collagen production, and adipose tissue health in the facial layers.
Materials / method: A controlled study was conducted by injecting DOT™ PN into the facial adipose and dermal layers of one side of the face, leaving the other side untreated as a control. Tissue biopsies were collected four weeks post-injection. Histological analysis focused on stem cell marker expression (CD31, CD34, CD166), fibroblast activation, collagen density, and adipocyte size and structure. AI-assisted image analysis quantified cellular and histological changes.
Results: DOT™ PN-treated areas showed a significant increase in CD31, CD34, and CD166 expression, particularly around blood vessels, indicating enhanced neovascularization and tissue regeneration. Fibroblast numbers and size increased markedly in the treated regions, contributing to collagen synthesis and dermal thickening. Adipocytes in the treated fat layer were larger, healthier, and exhibited reinforced cell membranes, demonstrating improved adipogenesis and fat layer integrity.
Conclusion: DOT™ PN injections significantly enhance the expression of stem cell markers (CD31, CD34, CD166), fibroblast activation, and collagen production, improving the structural and functional integrity of the facial dermis and adipose layers. These findings highlight DOT™ PN's potential as a powerful tool for skin rejuvenation, adipogenesis, and tissue regeneration in aesthetic medicine.
