Objectives: 1. The periorbital area – anatomy, histology, and aging pathogenesis.
2. Tixel mechanism of action (thermal mechanical fractional injury = TMFI) and advantages
3. Neocollagenesis and drug delivery - Comparison of EBDs.

Introduction: The periorbital region, comprising the delicate skin surrounding the eyes, is particularly prone to showing signs of aging, including fine lines, wrinkles, puffiness, and dark circles.

Many patients seek for non-invasive alternatives, that offer less downtime and minimal risk, and yet are effective. Among these, Tixel is safe and promising treatment modality for this area, offering optimal neocollagenesis and drug delivery.

Materials / method: A literature review was conducted to compare different EBDs usage in the periorbital area. This knowledge was combined with my practical experience with many energy based devices, including the Tixel.

Results: The periorbital area, surrounding the eyes, is a complex anatomical region, comprising multiple layers, including skin, subcutaneous fat, muscles, and underlying bony structures, its histology reflects its multifaceted function. The skin in this area is among the thinnest on the body, making it prone to visible signs of aging.

The Tixel can be applied safely into this region, heating the skin uniformly and open micro-channels for the delivery of essential active ingredients that synergistically improve periorbital rejuvenation potential.

Conclusion: The Tixel is optimal for periorbital area treatments, allowing individualization of the treatment and addressing all the pathologies associate with skin aging including wrinkles, texture, hyperpigmentation and skin laxity.

Objectives: - Investigate the mechanisms of cellular senescence in skin cells.
- Evaluate the impact of senescent cells on skin aging.

Introduction: Skin aging is characterized by the gradual loss of elasticity, the appearance of wrinkles, and a decline in the regenerative capacity of the skin. Cellular senescence is a major contributor to these changes. Understanding the role of senescence in skin aging is crucial for developing effective anti-aging therapies.

Materials / method: - Cell Culture: Human dermal fibroblasts cultured under standard conditions.
- Senescence Induction: Exposure to UVB radiation (30 mJ/cm²) to induce DNA damage-induced senescence.
- Senescence Markers: Analysis of SA-β-gal activity, p16INK4a expression by Western blot, and SASP factors (IL-6, IL-8) by ELISA.
- Cell lines: Human dermal fibroblasts
- Reagents: β-galactosidase staining kit, antibodies for p16INK4a, ELISA kits for IL-6 and IL-8
- Equipment: Incubator, centrifuge, spectrophotometer, Western blot apparatus

Results: Progerin expression -26.6%

Conclusion: Cellular senescence significantly contributes to skin aging through a dual mechanism of promoting chronic inflammation and causing structural degradation of the skin's extracellular matrix (ECM).

Although these senescent cells cease to divide, they remain metabolically active and secrete a variety of pro-inflammatory cytokines, chemokines, and proteases, a phenomenon known as the senescence-associated secretory phenotype (SASP).

Ex vivo study on human skin explants, cultured 10j, 4 applications (2mg/cm²) , immunostaining of progerin, image analysis for quantification of progerin level.

Results: Improved firmness +11% After 28 days / +23% After 56 days
Improved elasticity +17% After 28 days / +19% After 56 days

Conclusion: Aging is associated with inevitable accumulation of senescent calls.

Progescence and Pro-KL can slowdown cellular senescence both relicative senescence and premature or stress- induced senscence.

Cellular senescence is an irreversible state in which cells stop dividing. Excessive accumulation of senescent cells in tissue can adversely affect its regenerative ability, creating a pro-inflammatory environment conducive to the onset and progression of various age-related diseases. The skin is no exception to this observation either.
