Objectives: Specifically here we report about different in vitro experimental models, based on human cells.
Introduction: Hyaluronic acid (HA) is a naturally occurring biodegradable polymer with several biological applications in medicine including tissue engineering, dermatological fillers, and viscosupplementation for osteoarthritis treatment. Size dependent bioactivity has been recently approached in the scientific literature. Starting from the concept that the human skin has different layers and covers muscles and fat compartment, with a layer by layer approach we intended to analyse in vitro the potentialities of hyaluronan gels, and specifically of the novel High and low molecular weight HA complexes (HCC)
Materials / method: First: keratinocycets, fibroblasts and their co-culture. In addition 3D specific models were implemented for in vitro studies of the epidermal layer. Second: Adipose stem cells (ASCs) differentiation and proliferation were analyzed to establish biological activity of HCCs in subcutaneous compartment. Third: Mesenchymal stem cells, present in the stroma of the tissues, were used to study HCC effect on senescence. Finally, we are studying the HCC properties on muscle like cells. Results: Data showed that hybrid complexes proved to better promote wound healing of human keratinocytes in vitro respect to linear HA and supporting the clinically observed improvement of skin elasticity. In addition HCCs-based formulations clearly enhance adipogenic differentiation and proliferation, provide an appropriate microenvironment for MSC growth, by preserving their differentiation and senescence when compared with linear HA.
Conclusion: HCC proved to perform better than HA alone thus suggesting their potential as medical devices in aesthetic and regenerative medicine.
