Objectives: 1. To know that adipose stem cell exosome (ASCE) can be used as next generation regenerative therapeutics & aesthetics
2. To know that ASCE can reconstruct skin barrier functions and be a new therapeutic candidate for atopic dermatitis (AD)
3. To know that ASCE are toxicologically safe for skin
Introduction: Exosomes, nano-sized extracellular vesicles, are the most important mediator for intercellular communication. These days, ASCEs are being developed as cosmetics/cosmeceuticals and therapeutics as well. It was found that ASCEs can be very effective for skin rejuvenation/regeneration and atopic dermatitis through reconstitution of skin barrier functions. It was also found that ASCEs showed safe toxicological profiles.
Materials / method: Human adipose tissue-derived mesenchymal stem cell-derived exosomes (ASCE) were used for in vitro, ex-vivo, and in vivo studies.
Results: ASC-Exosomes,
1. contain active cargo: >1,000 proteins & ~600 miRNAs. Many of them are functional for skin;
2. Are effective for skin rejuvenation: increased collagen & elastin synthesis;
3. Are effective for atopic dermatitis in an inflammatory model: reduced clinical score, IgE level, cytokine mRNAs, & inflammatory cells;
4. Are effective for atopic dermatitis in a skin barrier model: reduced TEWL, epithermal thickness, & cytokines; increased the lipids; normalized gene expression for skin barrier, lipid metabolism, cell cycle, & immune; &
5. Are safe for skin: no toxicity was observed.
Conclusion: 1. ASCE can be next generation regenerative therapeutics and aesthetics, superior to conventional approaches such as skin boosters, growth factors, stem cells, stem cell conditioned media, PRP and others.
2. ASCE can be a novel promising cell-free therapeutic modality for AD treatment.
3. ASCE can be applied on skin with no significant toxicity.
