Frozen microneedle tray for delivering self-administrable botulinum toxin purified protein

Objectives: Solve pandemic challenges in the cosmetic industry, especially related to administration of botulinum toxin.
Self-administered prefilled BT tray comes to easily solve the challenges of traditional BT administration by providing for safe administration under remote medical guidance.
Storage of BT in prefilled frozen saline microneedle trays can prevent dilution errors and secondary side effects related to BT migration from injection site.

Introduction: The present innovative solution relates to a self-administered botulinum toxin purified protein through prefilled frozen microneedle trays. More specifically, it describes a disposable medical device used to administer transdermal quantitative units of botulinum toxin within specific dimensions of frozen 0.9% (preservative free) saline microneedle shaped trays. Due to pandemic business restrictions, patients were unable to receive medical procedures deemed as cosmetic. Thus, is needed a means of allowing application of BT without the need for a patient to be in a medical setting.

Materials / method: Different sizes and shapes of frozen microneedles, incorporating saline and BTPP can be grouped in a self-adhesive patch to be peeled of and applied to one targeted skin area. Each microneedle may include saline. The amount of preservative free saline used to build each of the microneedle to harbor e.g., 5 units of BTPP (the concentration can vary per microneedle and or tray depending on the target tissue and clinical applications) can and will be quantified to yield the right dilution (e.g., 0.5 ml). The microneedles containing lyophilized BTPP will mix and dissolve at target level.

Results: This system provides a unique method of prepackaging of BTPP in self-adhesive trays of frozen 0.9% saline as microneedles of different shapes and sizes and may be beneficial as compared to prior iced microneedles by providing a) a predetermined dilution of saline and BTPP per microneedle, b) a predetermined depth of penetration, c) most of the microneedle as opposed to just the tip may be dissolved in the target tissue, and d) the ability to provide for individual microneedle insertion or a patch line/group as prescribed.

Conclusion: Self-administered prefilled BTPP tray easily solve the challenges of traditional BTPP administration by providing for safe administration under remote medical guidance of patch like (peel off) frozen 0.9% preservative free saline microneedle trays filled with quantitative (2- 10 or more) units per microneedle. Prepackaging of BTPP into frozen microneedle trays with adhesive peel off patches with improved shape of the frozen medicated composite (no other carrier) allow for deeper skin penetration. The frozen saline and BT will mix and dissolve at target level(intradermal, subdermal, intramusc.)

Comparative analysis of human mesenchymal stem cells in respect to osteochondral tissue regeneration

Objectives: Congenital abnormalities, various diseases, and injuries may result in articular cartilage and bone degeneration. Recently, in the mentioned context, big hope is given to mesenchymal stem cell therapy. The main goal of the present study was the characterization and comparison of biological properties of MSCs obtained from adipose tissue (ATSCs), dental pulp (DPSCs), and urine (USCs) concerning bone and cartilage tissue engineering.

Introduction: Mesenchymal stem cells (MSCs) are generally characterized as undifferentiated cells with a unique ability of long-term self-renewing and plasticity. MSCs are adherent and have a fibroblast-like morphology. They are positive for a variety of surface markers, including CD29, CD44, CD73, CD90, and CD105. On the other hand, MSCs do not express markers of hematopoietic and endothelial cells. MSCs secern various bioactive molecules, which are involved in processes of regeneration. Due to these properties, MSCs represent a promising tool for tissue engineering and regenerative medicine.

Materials / method: MSCs from all tissues were cultured up to the third passage. They were morphologically analyzed by an inverted microscope and transmission electron microscope. The kinetics of proliferation was measured on days 1, 3, 5, and 7 via MTT assay. The expression of selected markers was assessed by flow cytometry. MSCs secretory profile was measured by Luminex MAGPIX® Instrument. Pellet cultures and a chondrogenic medium were used to induce chondrogenic differentiation. Osteogenic differentiation was induced by the osteogenic medium. The differentiation potential of MSCs was analyzed by real-time PCR.

Results: MSCs displayed similar morphology and had a similar course of proliferation kinetics. The highest rates were recorded in the case of USCs. MSCs shared the expression of typical markers, such as CD29, CD44, CD73, CD90, and CD105. They lack expression of hematopoietic and endothelial markers. In the case of USCs, interestingly the highest expression of SSEA was recorded. MSCs secerned similar levels of IFN gamma, IL10, IL18, and RANTES, while slightly differing in production of IL6, IL8, and MCP-1. MSCs had similar chondrogenic potential. On other hand, USCs had the lowest osteogenic potential.

Conclusion: In conclusion, it can be emphasized that MSCs from adipose tissue, dental pulp, and urine shared the majority of cellular characteristics typical for MSCs. They also displayed similar chondrogenic potential. On other hand, USCs had the lowest potential for osteogenic differentiation.

A case study of the difference in the patients condition and the degree of psychological stress reduction before and after DALA treatment

Objectives: Following DALA, the subjective degree of severity of the patient's facial condition
decreased, confirming the psychological change in the patient's self-perception of
improvement.
This survey confirmed the difference in the patient's condition and the degree of
psychological stress reduction based on self-perceptions of patients, independent of objective measures of the effectiveness of the treatment and changes in the patient’s face, and it was possible to predict the psychological satisfaction of the treatment.

Introduction: This study examines the difference in the extent of self-perceived facial problems in patients treated with DALA, a nonsurgical treatment for facial asymmetry and lifting.
In previous clinical studies of DALA, facial changes have been statistically significant when conducted at regular intervals and frequencies.
There has, however, been little research on the psychological satisfaction and factors determining the satisfaction of clinical treatment patients who choose this treatment because the changes are gradual and require continuous treatment, unlike other popular aesthetic treatments

Materials / method: Based on a survey of 11 patients who had received at least six sessions of DALA, this study analyzes and presents the differences between self-perceptions of their facial condition before and after treatment.

DALA treatments were performed on these patients using serine needles (0.14mm in diameter, 30mm in length), and needles (0.16mm in diameter, 30mm in length)
Treatments lasted 15 minutes and electronic acupuncture were performed using the STRATEK STN-330 model. No other medications or facial treatments were administered for 3 weeks.

Results: Questions 1 and 2 were asked to assess the change in the patients’ subjective perception of their facial condition before and after DALA.
The mean score for question 1 was 4.27, while the mean score for question 2 was 3.09, indicating a decrease of 1.18”

Questions 3 and 4 were asked to assess the change in the degree of psychological stress that patients who received DALA felt due to their self-perceived
facial condition before and after treatment.
The mean score for question 3 was 3.90, while the mean score for the question 4 was 2.18, indicating a decrease of 1.72”

Conclusion: Following DALA, the subjective degree of
severity of the patient's facial condition
decreased, confirming the psychological
change in the patient's self-perception of
improvement.

Moreover, the patient's subjective level of
stress decreased after treatment, indicating
psychological satisfaction with the treatment.

Animation deformity following breast augmentation or reconstruction: literature review and algorithm for prevention and treatment

Objectives: An algorithm for prevention and treatment of animation deformity was proposed.

Introduction: Animation deformity is a common complication following sub-pectoral implant-based breast augmentation or breast reconstruction. Various studies have stated the etiology and treatment of animation deformity. However, the algorithm for prevention and treatment has not been explored.

Materials / method: A literature survey was conducted in PubMed database using the keyword “animation deformity.” Studies related to definition, etiology, grading system, prevention, and treatment of animation deformity were collected.

Results: The etiology, grading system, prevention, and treatment of animation deformity were summarized. If mild or minimal animation deformity occurs, botulinum toxin or fat injection might be recommended. However, if moderate or severe animation deformity occurs, the patients should be advised to change the implant and reposition to pre-pectoral plane. If the patients are not willing to change the implants for pre-pectoral procedures, submuscular muscle splitting or selective nerve ablation can be recommended with or without botulinum toxins or fat injection.

Conclusion: Animation deformity is a common complication following submuscular breast augmentation or reconstruction that is difficult to treat.

An evaluation of a topical product containing blueberry extract and 30% Pro-Xylane in Chinese female with mild-to-moderate facial aging

Objectives: To evaluate the efficacy and satisfaction of a topical product containing blueberry extract and 30% Pro-xylane (test product) for improving facial aging in Chinese subjects with mild-to-moderate facial aging.

Introduction: Facial aging is characterized by fine lines around the eyes, mouth, and cheeks, skin laxity and lack of elasticity. The amounts of AGEs increase with aging and impact on each cell type in the body. Blueberry extract and 30% Pro-xylane can inhibit advanced glycation end products (AGE) formation and promote mucopolysaccharide production; both play an important role in facial aging. Currently, no clinical study has been published on the use of blueberry extract and 30% Pro-xylane in the Chinese population.

Materials / method: This prospective, randomized split-face, controlled trial enrolled 44 Chinese female participants with mild-to-moderate facial aging aged 30-60 years. One side of the face was randomized to receive the test product and the other side to be treated with the control product twice daily for 12 weeks. Transepidermal water loss (TEWL), skin elasticity, tightness, and dermal density were detected on both sides of the face at baseline and Days 28, 56, and 84 after enrollment. Clinical assessments were made by physicians and a satisfaction questionnaire was collected from each participant.

Results: Compared to baseline, the test product-treated side showed significant improvement in TEWL, skin elasticity, tightness, and dermal density at Days 28, 56, and 84. Compared to the control side, the test product-treated side showed better improvement in TEWL, skin elasticity, tightness, and dermal density at Days 28, 56, and 84. Clinical assessments showed similar results. Participants were completely satisfied with the outcome 84 days after using the test product.

Conclusion: The topical product containing blueberry extract and 30% Pro-xylane is effective in improving facial aging in Chinese women with mild-to-moderate facial aging.

Clinical evaluation of the moisturizing, soothing, and repairing effects of the Phyto Family regimen on sensitive skin

Objectives: The objective of this study was to prospectively evaluate the moisturizing, soothing, and repairing effects of the Phyto Family regimen on sensitive skin in Chinese patients.

Introduction: Sensitive skin is identified as a hypersensitivity reaction to stimuli, mainly on the face, involving symptoms such tightness, stinging, burning, tingling, pain and itching. Because of its complex etiology and pathogenesis, there is a lack of effective treatment. In the present study, we evaluated the benefits of a newly developed Phyto Family Regimen for the treatment of the multiple clinical signs and symptoms of sensitive skin.

Materials / method: A prospective clinical trial was conducted enrolling 66 Chinese women aged 18–45 years with sensitive skin. Participants applied the Phyto Family regimen, a combination of Phyto Serum, Phyto Mist and Phyto consisting of hyaluronic acid and botanical ingredients, to the whole face. The skin redness values a*, moisture content of the stratum corneum, the transdermal water loss, and skin glossiness were measured at baseline and 15 minutes,30 minutes and 1 hour after using the Phyto Family regimen. Clinical assessments by physicians and satisfaction questionnaires from participants were collected.

Results: Compared to baseline, the skin showed a significant decrease in a* redness values and transdermal water loss, and a significant increase in the moisture content and glossiness of the cutaneous stratum corneum after using the Phyto Family regimen (Figures 1–4). The clinical assessment of skin redness was partially improved in 78.79% and significantly improved in 16.67% of the participants. The participants were 100% satisfied with the treatment after 1 hour of using the Phyto Family regimen.

Conclusion: The Phyto Family regimen achieves satisfactory moisturizing, soothing, and repairing effects on sensitive skin.

Is upper medial canthus compression enough during filler injection in glabella to avoid retrograde filler embolism?

Objectives: While many studies suggest compressing the upper medial canthus region with a non-dominant finger while injecting filler in the vertical wrinkles of the glabella region, we investigated by ultrasound how effective the compression is in this region in avoiding retrograde embolism.

Introduction: Hyaluronic acid (HA) fillers have been widely used for reversing the signs of the aging face. Injectable fillers are particularly effective in treating vertical glabellar wrinkles. One of the dangerous complications of this injection is blindness

Materials / method: 165 clients of an Aesthetic clinic from September 2022 to November 2022 were randomly subjected to 20MHz high frequency power Doppler imaging . First, ultrasound was performed transversely in the glabella area to determine the prevalence of central and paracentral arteries. In cases where the artery was within one centimeter of the midline of the glabella region, simultaneously with the placement of the probe transversely, the area above the Medial canthus was compressed with the index finger(figure2). Results: In 48 cases (29%) of 165 cases, the artery was located within 1 cm of the midline of the glabella region. One artery was observed in 23 cases (13.9%), two arteries in 15 cases (9%), three arteries in 7 cases (4.2%) and four arteries in 3 cases (1.8%). In all the cases where the artery was within 1 cm of the midline, only in 60% of the cases, by compressing the upper part of the internal canthus, the blood flow of the glabella region was stopped in Power Doppler.And in all cases, by compressing the upper part and inside the medial canthus, blood flow was not observed in the glabella region.

Conclusion: The results of these observations suggest that the compression of the upper part of the medial canthus alone is not enough to avoid retrograde embolism, and it is better to also compress the inner part of the medial canthus during filler injection in the glabella region.

How to turn your wound healing from zero to hero using skin substitute grafts

Objectives: The objective of this study is to overview common and/or popular skin substitute grafts, what they are made of, how they are processed, and benefits to each type in terms of wound healing.

Introduction: Chronic wounds such as wounds secondary to skin cancer removal, diabetic foot ulcers, and pressure ulcers are notorious for taking a long time to heal. It has been shown that skin substitute grafts are able to promote wound healing. This abstract will briefly overview the three common skin substitute graft types used in clinics.

Materials / method: No materials were used in the making of this abstract. My data and information was gathered from the cited sources in the reference section.

Results: No clinical data was gathered for this poster. Main objective is to spread awareness of three different types of skin substitute grafts used in dermatology offices that can promote wound healing.

Conclusion: These three skin substitute grafts have been clinically proven to increase rate of wound healing as well as percentage of wounds completely healed. These are great options for clinics that deal with chronic wounds.

Current status and future directions of stem cell therapy in burn injuries: a systematic review

Objectives: Studies using animal models have shown promise for stem cell therapy in accelerating wound healing, whereas a critical evaluation of the available research is still necessary. Therefore, we conducted a thorough assessment of the experimental and clinical data available to date to evaluate the potential of stem cell therapy in treating burn wounds.

Introduction: Stem cells are unique due to their ability to regenerate and differentiate into different cell types, providing hope for treating diseases without effective treatments. Stem cell therapy for wound healing, particularly for burns, is being investigated as it remains a significant global health issue. Although burn therapy has advanced, achieving complete wound coverage and functional skin regeneration remains challenging. Stem cell therapy has shown promise in animal studies for speeding up wound healing, but further critical analysis of experimental and clinical data is necessary to assess its

Materials / method: Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) statement was used to identify relevant titles and abstracts from multiple databases. Non-English articles, conference papers, case studies, and reviews were excluded from the search. In addition, a manual search was performed by reviewing references from other papers and books. After a thorough evaluation, 20 articles met the eligibility requirements for inclusion in the study.

Results: Various experiments were conducted using different animal models, sources, dosages, and administration routes. Out of the 21 studies reviewed, 16 reported positive outcomes of stem cell therapy on these variables, suggesting its potential for improving burn wound healing. Stem cell therapy has been found to increase collagen levels, promote angiogenesis by increasing blood vessel count and levels of pro-angiogenic factors such as VEGF and angiopoietin, exert anti-inflammatory effects, enhance tissue viability, and reduce cell death in burn wounds compared to controls.

Conclusion: Stem cell therapy can potentially improve burn wounds and provide systemic anti-inflammatory effects. However, the limitations of this research, including a small number of trials and distinctive variables, should be recognized. Further study is required, particularly in larger animal models and clinical trials.
