From stem cells and exosomes to cracking proteins’ amazing structures
Цели: The antiaging discovery process focuses on scientifically smaller molecular mechanisms, shifting from stem cells that contain exosomes, to exosomes, which contain proteins, to proteins, which are the brains of the cells and the most important determinant factors in antiaging medicine. The future appears bright, but there are several unknowns about the side effects of constructing new proteins. Additionally, these three agents depend on systemic signalling resonance to refold misfolded proteins and transport stem cells and exosomes to their site of action
Введение: Discuss the popularity of stem cells, their benefits, advantages and disadvantages. A review of 250 articles reveals that the of the most challenging aspect of incorporating stem cells into clinical practice is controlling their division and differentiation potentials. A review of over 300 articles examines exosomes' advantages and disadvantages. It delineates the complexity and often miraculous, yet controversial effects of exosomes. A review of 120 articles highlights he future where AI will be used to design and produce new proteins
Материалы / методы: The Past: Stem cells offer tantalising prospects to anti-ageing and regenerative medicine. They are used for skin repair, hair growth and the maintenance of various adult tissues and organs. They can modulate numerous incurable diseases such as heart conditions, diabetes, brain injuries, etc. The necessary process for stem cells functionality, cellular differentiation, depends on the increased ratio between:
1/ mitochondrial differential that promotes activity, and
2/ nuclear differentiation that prevents activity.
Another caveat in this process is immunorejection.
Результаты: Incapacitating the immune system with antibodies that block normal T-cell activation, even temporarily, may win the battle over immunorejection but lose the war over tumours growing unobstructed since the immune soldiers are paralysed. The present: Exosomes regulate complex intracellular pathways and have been used as biomarkers, cell-free therapeutic agents, drug delivery carriers, exosome kinetics, and cancer vaccines. The complex cargo of exosomes is readily accessible via sampling of biological fluids (liquid biopsies).
Заключение: Proteins, metabolites, and nucleic acids delivered by exosomes into recipient cells effectively alter their biological response. Such exosome-mediated responses can be healing or the exact opposite: They can promote disease and/or aging. The future: The Nobel Prize in Chemistry 2024 is about proteins, life’s ingenious chemical tools. David Baker has succeeded with the almost impossible feat of building entirely new kinds of proteins. Demis Hassabis and John Jumper have developed an AI model to solve a 50-year-old problem: predicting proteins’ complex structures.
