Objectives: We demonstrate tissue growth in subcutaneous injection of plant derived rhCollagen gel in rat model. Cell proliferation and angiogenesis was evident within the injected gel committed which coincided with natural sustained release of growth factors including vasculo-endothelial growth factor (VEGF) and platelet derived growth factor (PDGF) in a course of up to 45 days. We have tested different rhCollagen/HA formulations that displayed constant and low injection force even when injected via 32G needle, indicating ease of use for accurate deployment of the gel at selected sites.
Introduction: Hyaluronic acid have proved itself as a good dermal filler. However, its limited ability to promote cell proliferation and angiogenesis result in limited tissue integration which may cause face distortion, particularly in extreme face expressions. Collagen comprises the major component of connective tissues. However tissue extracted collagen suffer from many disadvantages, including inconsistency immunogenicity and risk of disease transmission. We are producing clinically tested human recombinant collagen in tobacco plants.
Materials / method: Plant derived rhCollagen was fibrillated and cross-linked by carbodiimide. Female Sprague Dawley rats were injected with 0.5 ml of rhCollagen gel at the dorsal plane of the rat’s back. Animals were sacrificed at time-points 1, 7, 14, 21, 30 and 45 days post-treatment and implants were assessed by histopathology and PDGF and VEGF content by ELISA. rhCollagen/HA formulations were prepared and injection force was tested by injection via 32G needle

Conclusion: Plant derived rhCollagen promote tissue regeneration and holds great potential to augment the performance of HA derived dermal fillers by promoting tissue integration. Furthermore, rhCollagen maybe used as a matrix for PRP as already have been proven clinically and possibly for stem cells from different source in tissue augmentation.
