Advanced autologous injectable platelet derived exosomes rich plasma and final product characterization (educational grant from D-MED)
目标: Exosomes are small extracellular vesicles ranging from 50-200 nm in diameter, playing a crucial role in cell communication, tissue repair, and regeneration. In these collective studies an innovative therapy method based on Platelet Rich Plasma (PRP) to induce the release of billions of vesicles by stressing platelets and other blood cells within PRP. Through specialized preparation protocols, this method enhances platelet activation, providing a plasma rich in exosomes derived from platelets. This approach finds extensive applications in aesthetic, orthobiological, and therapeutic treatments
介绍: This study aims to develop an advanced PRP variant targeted for aesthetic and medical applications. The treatment method delivers effective outcomes in various areas such as skin regeneration, hair loss, spot treatments, and joint and connective tissue therapies. Notably, this method is entirely autologous, utilizing exosomes derived from the patient's own platelets. This approach ensures safety by being closed to horizontal gene transfer, thereby eliminating the risk of immune reactions or other complications associated with foreign biological materials.
材料 / 方法: Using a specific 600µm filter-blades were employed to stress the cells, inducing the secretion of billions of vesicles and promoting platelet activation. The shear stress created by the blades applied mechanical forces to the platelets, leading to their activation and subsequent exosome production. A vacant tube was used to further facilitate the secretion of billions of vesicles by cells and the activation of platelets. This tube generated vortices through varying G-forces, continuing the activation process.
结果: CD9 (Platelet): 99.6% • CD45 (Pan Leukocyte Marker): 0.3% • CD41 (Platelet GPIIb, IIb Integrin): 99.4% • CD61 (Platelet Glycoprotein GPIIIa): 98.6% • CD81 (Target for Anti-proliferative Antigen 1 [TAPA-1]): 98.5% • Viability: 98.9% These CD marker percentages help us determine the origin of the vesicles. The high levels of CD9, CD41, CD61, and CD81 confirm that the majority of the vesicles are exosome-derived from platelets. The low percentage of CD45 further supports that the vesicles are not derived from leukocytes, reinforcing the purity and targeted effectiveness of our method.
总结: Due to its autologous nature, this technique poses no risk of allergic reactions post-injection, making it a safe and effective treatment option in aesthetic and medical applications.
