Objectives: Here, we present a new skin depigmenting compound isobionic-amide, as a new inhibitor of tyrosinase and with potentiating effect on tyrosinase inhibition by cysteamine. The results of in vitro and clinical studies of isobionic-amide cysteamine formula will be presented.

Introduction: Due to the suboptimal efficacy and safety of current skin depigmenting compounds, there is a continuous need for new safe and efficacious depigmenting formulations. Cysteamine has been shown to be a potent melanogenesis inhibitor, due to its inhibitory effect on tyrosinase and peroxidase, the two main enzymes in melanogenesis. Several clinical studies have assessed the safety and efficacity of stabilized cysteamine for the treatment of hyperpigmentary disorders

Materials / method: The effect of isobionic-amide, cysteamine and their combination on tyrosinase inhibitory activity was determined.
The efficacy and safety of topical isobionic-amide cysteamine combination were evaluated in two double-blind randomized trials, one in comparison with vehicle in 40 patients with PIH, the other in comparison with placebo and modified Kligman’s formula in 80 melasma patients. Chromametry, total post-acne hyperpigmentation index (TPHI) or modified melasma severity index (mMASI), and qualitative measurements were collected at baseline, week 4, week 8 and week 16

Results: Isobionic-amide and cysteamine were both inhibitors of tyrosinase. The combination of both molecules had a synergistic effect on the reduction in tyrosinase activity (67% inhibitory effect).
In PIH patients, all parameters (melanin index, TPHI, quality of life) showed significant improvement in isobionic-amide cysteamine group versus vehicle.
In melasma patients, the isobionic-amide cysteamine complex and Kligman’s formula significantly reduced melasma severity, with an equivalent onset of action of 4 weeks and ending with a comparable efficacy at week 16.

Conclusion: Isobionic-amide synergistically enhanced the tyrosinase inhibitory effect of cysteamine. Isobionic-amide Cysteamine complex was significatively more effective than placebo in the treatment of PIH. Isobionic-amide Cysteamine had the same efficacy as the Kligman’s formula for the treatment of melasma with the same onset of action and shows superior improvement in quality of life and patients satisfaction. The data suggest that the Isobionic-Amide Cysteamine could potentially be a novel alternative to modified Kligman's formula for the treatment of hyperpigmentary disorders.

Objectives: 1-To assess the effectiveness of peptides in reducing skin wrinkles compared to placebo in randomized controlled trials (RCTs).

2-To evaluate the impact of peptides on skin hydration, elasticity, roughness, brightness, texture, and density.

Introduction: Skin aging is characterized by the development of wrinkles, loss of hydration, decreased elasticity, and other structural changes. Peptides, bioactive molecules composed of short amino acid chains, have been increasingly investigated for their potential role in reducing these aging-related alterations. We aim to evaluate the efficacy of peptides as anti-aging agents on key dermatological parameters, including wrinkles, skin hydration, elasticity, roughness, brightness, texture, and density.

Materials / method: A thorough systematic review and meta-analysis was conducted in accordance with PRISMA guidelines. We employed a random-effects model based on the DerSimonian and Laird technique, using the inverse variance method. Forest plots were generated to illustrate the results, including direct comparisons between different types of peptides and their modes of application. Heterogeneity was assessed using Cochran’s Q test, with a significance threshold of p < 0.005, and I².

Results: Peptide treatment resulted in a statistically significant reduction in skin wrinkles, with a mean difference of 0.3518 (95% CI: -0.0021 to 0.7057, p = 0.0514). Subgroup analyses demonstrated that polypeptides exhibited the most pronounced effect on wrinkle reduction (MD = 0.9083) compared to placebo (MD = 0.1837). Peptides also significantly improved skin hydration (MD = 5.7971, p < 0.0001), with tripeptides showing the greatest increase (MD = 16.5). The impact on skin elasticity was less marked, with a non-significant mean difference of 0.0996 (p = 0.1536).

Conclusion: Peptides, particularly polypeptides and tripeptides, demonstrate significant efficacy in reducing wrinkles, improving skin hydration, and enhancing brightness, with lesser but positive effects on skin elasticity, roughness, and texture. To conclude, peptides represent a promising intervention for age-related skin changes, though further high-quality studies are needed to clarify their long-term effects and optimize their application in dermatological practice.

Materials / method: This study investigates clean beauty ingredient safety through a comprehensive literature review. Key databases such as PubMed, MEDLINE, and Embase were systematically searched using targeted Medical Subject Headings (MeSH) and keywords. Peer-reviewed articles, clinical trials, and reviews were meticulously selected for their relevance and recent publication dates. Data extraction and synthesis focused on evaluating the scientific evidence behind commonly scrutinized ingredients, ensuring a rigorous analysis of their safety profiles.

Results: Irritants: MI, fragrance, and formaldehyde are confirmed allergens linked to dermatitis, validating their exclusion in clean beauty.
Endocrine Disruptors: The link between chemicals like Triclosan and hormonal disruption remains uncertain, with most studies conducted on animals.
Carcinogens: Formaldehyde and 1,4-dioxane are associated with cancer risk, though evidence is still emerging.
Overall: Clean beauty promotes scrutiny of ingredients, but more research is needed to fully validate the safety of many claims.

Conclusion: The clean beauty movement has highlighted the importance of scrutinizing cosmetic ingredients, revealing that some commonly avoided chemicals, like MI, fragrance, and formaldehyde, are linked to known health issues. However, the safety of many other "clean" ingredients remains inconclusive, with limited human studies. While eliminating certain harmful ingredients is prudent, more research is needed to fully understand the effects of many "clean" cosmetics components on human health.
