Objectives: Nutraceuticals – orally taken supplements derived from botanicals – may interfere with aging, in a multifactorial gut-brain connecting way that then turns out to be beneficial for delaying skin health and beauty decay. Chasing nutrients consumed daily for anti-aging effects, we came across a rare botanical genus eaten in daily broths in Asia as early as 2000, the small molecules of which seem able to reverse cell senescence. Based on their properties to inhibit IL-6, TNF- and rho A mediated cell signaling, we named them AISA for “Anti-Inflammatory Senescence Actives”.
Introduction: In recent years, small molecules have shown their value in the treatment of auto immune diseases and also being associated in co-treatment of infectious diseases. Their mechanism of action is based on their capacity to act as Jak-inhibitors as well as rho cascade inhibitors. By our bio-molecular approaches they turned out to be the solution of the challenges of aging progression – when the consequences of a generalized silent inflammation may accelerate the cell senescence process – itself entertaining the loop of an ongoing chronic inflammation.
Materials / method: The RISTOMED EU FP7 phase 2A multi-centric first in human clinical trial has validated the clinical efficacy of AISA monoterpenenoids as geroprotectors, acting as biologics and also reducing stress, strongly down-regulating pro-inflammatory cytokines via inhibition of NF-B, as well as the thrombosis mediator fibrinogen, but also metabolic markers such as insulin and HOMA-IR.
Results: Terpenoids have been recently acknowledged as geroprotectors and monoterpens as biologics in countering cytokine storms of various origins, metabolic or infectious. Chronic inflammation is the common ground for accelerated aging, also enhancing SASP (Senescence Associated Secretory Phenotype). Previous results of our studies also include interesting results on neuro-active potential of microbiota and the repair of gut barrier repair and thus the mucosal immunity.
Conclusion: Small molecules such as the mono-terpene d-Limonene, should be considered valid candidates for issues relevant as preventing Common Chronic Diseases (CCD) enhancing healthspan and longevity. Yet, thanks to the gut-brain axis that actively communicates with the skin, they turned out to greatly contribute to the conservation of youthful properties, mainly based on the anti-inflammatory effect, especially in those psycho-dermatologic syndromes such as Atopic Dermatitis or Psoriasis.
