Objectives: The use of adipose tissue products is constantly growing as well as the development of new devices for their processing. The benefit brought to regenerative practices has gone far beyond the boundaries of specialties, thus much more attention is given to understanding the mechanism behind.
Such a rapid growth could not be accompanied by an adequate organization of knowledge, a standardization of procedures, no mentioning the development of clear guidelines, with the result of an easy confusion about, and the consequent limitation in findings.

Objectives: The aim of the project consisted of the control of viability and phenotype of adipose derived cells using a guided procedure with the two types of cannulas having 0.8 mm and 1 mm side port holes and processed following SEFFI (Superficial Enhanced Fluid Fat Injection) technique; the results were compared with tissue harvested with a standard liposuction technique and processed with a standard procedure consisting of enzymatic digestion (collagenase). .

Introduction: Injection of autologous adipose-derived stem cells (ADSCs) and a stromal vascular fraction (VSF) into dermal and subdermal layers promises regenerative advantages by improving skin volume and rejuvenation. It's extremely important to set a Good Protocol (GP) to harvest and manipulate the adipose tissue in order to achieve the higher cell viability and the highest amount of SVF cells and ADSCs.

Materials / method: This study was performed on adipose tissues harvested from 7 patients (6 females and 1 male) with an average age of 48.5 years with 3 different techniques.
Adipose tissue using a 0.8- and 1-mm cannula were monitored for their cell viability using absorbance spectroscopy ; a.t. harvested using Coleman was processed by enzymatic treatment as quality control of tissue. For phenotype analysis Cells were incubated with antibodies anti human against CD90-FITC, CD73-PeCy7, CD44-PE, CD31-PECy5, CD235a- PECy7, CD34-FITC, CD45-FITC and CD146-FITC and read using the flow cytometer .

Results: Lipoaspirate tissue derived by 0.8- and 1 mm cannula from all samples proved to be vital and possess viable cells.Average of the absorbance was similar immediately after plating (t0) and 72 hours after (t72) for the two cannulas, 0.8- and 1 mm cannula. The two systems proved to equally harvest vital tissue.
Morphologically, cells harvested by different cannula or isolated by enzymatic digestion showed differences in their morphology.

Conclusion: The results proved the vitality of adipose tissue harvested using the different cannulas.Tissue was vital in both cannulas and the values were comparable to the isolation of stromal vascular fraction (SVF) by enzymatic digestion, the gold standard procedure for adipose tissue derived cells. Cells to be defined mesenchymal stem cells, need to adhere to plastic surface and express specific surface antigens, especially CD44, CD90 and CD73.Cells derived from each individual were able to attach and proliferate for at least three passages in culture and show the typical fibroblastic morphology.

Objectives: To evaluate the efficacy of NCES prepared in SVF compared to the laboratory-based method of NCES preparation in PBS.

Introduction: Stromal vascular fraction (SVF ) is a very rich source for mesencymal stem cells, pericytes and other important stem cells important for pigmentation, wound healing and regeneration. Non cultured epidermal cell suspension has revolutionized the treatment of stable vitiligo. Prepration of NCES in SVF (replacing prosphate buffer saline {PBS} in final solution).

Materials / method: Anatomically based matched lesions (26 pairs) in 20 stable vitiligo patients were randomized to receive NCES prepared in PBS or SVF. Each patient was evaluated at 4, 8, 16 weeks after surgery by a blinded observer with regard to extent of repigmentation, colour match, patient global assessment (PGA) and pattern of repigmentation.

Results: Repigmentation outcome in SVF method as compared to PBS NCES was: excellent (≥90%) repigmentation: 42% vs. 35%, p=1.000, good (≥75%) repigmentation : 75% vs. 70%, p=1.000; colour match: 64% vs. 54%, p=0.784, patient satisfaction based on PGA score – 24.02 vs. 23.39 (p=0.220) and major pattern of repigmentation (diffuse) – 79% vs. 69% (p=0.628).

Conclusion: Preparation of NCES in SVF is an effective, superior and regenerative technique for vitiligo surgery in clinical practice.

Objectives: 1: Determine sarcopenia as the cause of muscle atrophy
2: Formulate ideas for processing adipose stem cells in an in vitro model of dexamethasone-induced myotube atrophy

Introduction: Sarcopenia is a cause of muscle atrophy. The prevalence of sarcopenia increases with age, ranging from 5-13% in 60-70 years old to 11-50% in people over 80 years old. The literature confirms that sarcopenia is associated with adverse clinical outcomes. Therefore, how to treat patients with sarcopenia in later stages or those who fall into sarcopenia because of medical treatment, develop drug therapy to increase muscle mass and strength, improve patients' quality of life and survival rate, is an urgent but not yet received met medical needs.

Materials / method: In this study, we assessed the effect of ADSC on muscle differentiation and muscle atrophy in vitro through undifferentiated C2C12 muscle cells, by observing cell morphology, immunofluorescence staining and using qPCR. To evaluate whether ADSCs has the ability to promote the differentiation of mouse skeletal muscle cells, and to use Dexamethasone to induce atrophy of differentiated C2C12 myotube cells, so as to establish a cell model of muscle atrophy. By observing the morphology of muscle cells and using qPCR to detect and analyze the changes in muscle.

Results: According to past research, stem cells have the function of tissue regeneration, and stem cell transplantation is an important way to improve muscle atrophy caused by sarcopenia.

Conclusion: Stem cell therapy technology is gradually moving from basic research to clinical application. This project presents the impact of ADSCs in sarcopenia causing muscle atrophy.

2. Equal harvesting attempts made on each square by using a 0.85 mm sharp motorized punch
3. Micro -Pictures of wounds were taken with high resolution USB camera immediately after the extraction and 24 hours later in both cases

4.Injection of 0,2ml PRP only within the area A

5. The surface size of wounds in each rectangle was measured using a proprietary image processing system.

Results: All the data from all 49 wounds
analyzed by applying independent sample t-test and it was found that p=0.000<0.05.
The mean surface of the wound, without PRP , reduces by 21.3 %, while the mean value of the wound with PRP reduces by 49,95 % within the same time frame .

The decrease in wound surface with PRP is 134.5% larger than that without PRP

Conclusion: The injection of PRP plays an extremely important role in accelerating the healing process in the donor area thus improving patient comfort

Materials / method: Materials & Methods: We performed the treatment on 50 patients with androgenetic alopecia. The total number of sessions was 4 every month. Researcher clinical evaluation, global photography and trichoscopy with measurement of the Density / cm2 ratio by trichoscopy were collected at every session of therapy. All patients filed out a brief questionnaire of self-assessment.

Results: Results: Results of our study were very promising, with improvement in most of patients seen with both global photography and trichoscopy. All patients defined the treatment as “painless and pleasant” and all were satisfied of the clinical result.
